Does anyone know how these are different from the old snug and dry and natural care versions? Thanks!

I was curious too, so I googled it like crazy last week but found nothing. I found no retailer besides Costco having the "plus" and it's not even on the Huggies website.
I therefore think that the "plus" graphic on the boxes simply means the quantity is higher. Huggies probably makes their huge-quantity boxes available only a bulk warehouse retailers like Costco.
I see what I think is your point, though - the cute "plus" graphic in the middle of the box (not in the corner where the quantity is noted) does imply a product upgrade rather than a higher quantity. But if it was an upgrade Huggies would undoubtedly say something about that on their website.

Extra capacity to hold pee, or to put it more delicately, more absorbent than regular Snug and Dry. Thicker than a regular diaper but quite a nighttime diaper. They are great for my DS, I love them.
